Se le reconoce como el organizador de los fotoperiodistas encargados de documentar la "Gran Depresión" para la oficina de Administración de Seguridad Agraria. Perteneció a infantería durante la Primera Guerra Mundial y al finalizar realizó estudios de Economía en la Universidad de Columbia, donde después sería profesor. En ella coincidió con el profesor Rexford Tugwell que cuando fue nombrado Subsecretario de Agricultura puso en marcha la Resettlement Administration que posteriormente pasó a denominarse Farm Security Administartion (FSA). Ese nombre lo mantuvo durante siete años, ya que al iniciarse la Segunda Guerra Mundial, en 1943, sus recursos se incorporaron a la Oficina de Información de Guerra. Al clausurarse en 1943 arrojó un balance final de 180000 negativos, 1500 diapositivas en color y más de 100000 positivos dotados de comentario escrito. Aunque comenzó realizando fotografías para Resettlement Administration, pasó a ocuparse de la dirección de la sección histórica, desde donde planteó la creación de un registro fotográfico para plasmar los efectos de la depresión. El tipo de fotografías que quería debían cumplir una doble función, por un lado ser informativas y por otro que fuesen también simbólicas de la situación de sequía y tormentas de polvo. De esa forma se trataba de dar una visión de Estados Unidos a los estadounidenses en el marco del New Deal y con planteamientos propios del realismo social. La selección de fotógrafos encargados de esa tarea fue de gran relevancia. Entre ellos se encontraban Walker Evans, Dorothea Lange, Gordon Parks, Margaret Bourke-White, , , , Ben Shahn, John Collier, , , Jack Delano, Russell Lee, , Arthur Rothstein, , Marion Post Wolcott o Willard Van Dyke. En 1973 publicó sus experiencias en el artículo The Farm Security Administration Collections of Photographs, con Nancy Wood. Al terminar la Segunda Guerra Mundial entró a trabajar para Standard Oil donde organizó un equipo de fotógrafos para dar a conocer la empresa; entre ellos se encontraban Berenice Abbott, Gordon Parks, , Esther Bubley, , Russell Lee, , Elliott Erwitt y . Algunos le siguieron posteriormente a un nuevo proyecto en Pittsburgh, la Pittsburgh Photographic Library. En 1960 su colección de fotografías se transfirió a la Carnegie library. (es)

He headed the Information Division of the Farm Security Administration (FSA) during the Great Depression, and launched the documentary photography program of the FSA. It hired photographers to travel across the United States and document people in different areas and settings as part of showing the state of people in rural areas in those years. Specific projects were conceived to help assess effects of government programs. He later worked several years on a documentary project for Standard Oil, established the Pittsburgh Photographic Library (PPL), consulted with other companies, and taught photo-journalism at University of Missouri. In his later years he returned to the West, living at last in Colorado. (en)

Efter tjänstgöring i USA:s armé under första världskriget utbildade sig Roy Stryker till ekonom på Columbia University i New York. Han använde fotografier för att illustrera sina facktexter och föreläsningar. Han arbetade på universitetet tillsammans med Rexford Tugwell, och när denne anställdes av Resettlement Administration, senare omdöpt till Farm Security Administration, följde Stryker med honom dit. Tugwell och Stryker tog initiativ till att Resettlement Administration skulle dokumentera jordbruksområdenas problem under Den stora depressionen och 1935 blev Stryker chef för myndighetens informationsavdelning, där han organiserade ett fotodokumentationsprojekt. Roy Stryker ledde detta projekt med fast hand och såg till att de stora tidningarna och tidskrifterna fick tillgång till fotografierna. Detta underlättade att allmänheten fick kännedom om problemen för de fattiga på landsbygden och ledde också till att de engagerade fotograferna gjorde kommersiell karriär. Över en knapp tioårsperiod tillgängliggjordes för pressen 77.000 fotografier från 164.000 negativ. Roy Stryker engagerade som fotografer bland andra Dorothea Lange, , Walker Evans, Ben Shahn, , , , Jack Delano och Gordon Parks. Under andra världskriget knöts myndighetens fotografienhet till Office of War Information och fick mer propagandistiskt uppdrag under ett och ett halvt år, innan den upplöstes. Vid samma tillfälle lades Food Security Administration ned, varefter dess fotosamling överfördes till Library of Congress. Roy Stryker övergick till att arbeta inom Standard Oil med dess PR-dokumentärprojekt 1943–1950. Fotografer för Standard Oil-projektet var bland andra Berenice Abbott, Gordon Parks, , Russell Lee och , varav de tre senare följde med Stryker till dennes nästa projekt i Pittsburgh i Pennsylvania. Efter att ha angivit ämnen, som han ville ha belysta, lämnade Stryker fotograferna fria händer att genomföra fotograferingen efter egna infallsvinklar. Mellan 1950 och 1952 arbetade Stryker med att bygga upp Pittsburgh Photographic Library, vars samlingar 1960 överfördes till Carnegie Library i Pittsburgh.
