He is best remembered for his contributions to mathematics, his membership in the Scriblerus Club (where he inspired both Jonathan Swift's Gulliver's Travels book III and Alexander Pope's Peri Bathous, Or the Art of Sinking in Poetry, Memoirs of Martin Scriblerus, and possibly The Dunciad), and for inventing the figure of John Bull. Sus mordaces escritos contienen cierta alegoría política, como The History of John Bull de 1712, que instauró al personaje John Bull como una personificación nacional de Inglaterra. Junto a Alexander Pope y Jonathan Swift, Arbuthnot fue miembro fundador del célebre Club Scriblerus, que "congregaba hombres de letras del partido Tory y habitualmente enfrascados en violentas polémicas contras los Whigs." Entre otras actividades, este club tenía el propósito de ridiculizar la mala literatura y el aparente aprendizaje. (es)

Arbuthnot bezocht het Marischal College in Aberdeen en ging in 1692 naar Oxford om medicijnen te studeren. Hij behaalde zijn MD aan de Universiteit van St Andrews in 1696.
